Extraction of polycyclic aromatic hydrocarbon (PAHS) from complex matrices like soil samples does not always give accurate and reproducible results. This has led to the suggestion of several extraction techniques like Soxhlet, ultrasonication, mechanical shaking e.t.c. However, some of these techniques suffer from draw backs like long extraction time, loss of analyte due to reconstitution in new solvents, permanent adsorption of analyte to the adsorbent in course of clean-up. In this study, comparison of some extraction methods and clean-up procedures for the 16 priority USEPA PAHs were investigated with the aim of determining the best technique for quantification of PAHs in soil. Soil samples were extracted using three methods (Soxhlet, ultrasonication and mechanical shaking) and the extracts were further cleaned using three techniques: packed columns (silica gel packed), membrane filter and solid phase extraction with polymeric sorbent. The extracts were analysed for the 16 USEPA PAHs and comparison made of these techniques. Quantification and identification was made with gradient elution using acetonitrile and water in a reverse phase high performance liquid chromatograph (HPLC) with a UV detector. The results showed that ultrasonicationusing a membrane filter gave the highest extract efficiency as well as having the advantage of shortest extraction time and lowest solvent consumption.en_US
