- ItemOpen AccessAntioxidant Enzymes and Lipid Peroxidation in Alloxan-Induced Diabetic Rabbits(Science Publishing Group, 2018-06-15) Enefe, N.D.; Ebuehi, O.A.T.The study was carried out to evaluate the effect of alloxan–induced diabetes on some oxidative stress markers in rabbits. Ten rabbits weighing between 1.0-1.5kg were used and divided into two groups of five rabbits each. Diabetes was induced in one of the groups, by intraperitonial administration of alloxan at a dosage of 150mg/kg body weight and the control rabbits were administered normal saline (0.9% NaCl). The animals were monitored for 14days after confirmed diabetes in the test group. The fasting plasma glucose, plasma insulin, plasma lipids, the antioxidant enzymes Superoxide dismutase (SOD), catalase, and lipid peroxidation were assayed for, in the plasma and the brain of the diabetic and non-diabetic rabbits. The results showed a significant decrease in mean body weight of the alloxan-induced diabetic and control rabbits pre and 14days post alloxan-induction. There was a significant (p < 0.01) increase in fasting plasma glucose, a significant (p < 0.01) decrease in the activity of SOD and a significant (p < 0.01) increase in the activity of catalase in the plasma and brain; there was also a significant (p < 0.01) increase in the level of lipid peroxidation, plasma cholesterol and triglycerides in the diabetic rabbits. Culminating a depletion of SOD activity, the altered activity of plasma (or brain) catalase and increased lipid peroxidation isdue to the oxidative stress induced by the diabetic condition, hence the present study indicate that alloxan – induced diabetic potentiated oxidative stress in a short term duration. This shows the importance of antioxidant mimetics to be administered at early stage of diabetics in other to increase the antioxidant capacity thus preventing oxidative stress which may lead to complication in diabetes.
- ItemOpen AccessEvaluation of the Therapeutic Effects of Aloe Vera Gel on Alloxan- Induced Diabetic Rats(Nigerian Quarterly Journal of Hospital Medicine, 2016) Imaga, N.O.A.; Ebuehi, O.A.T.; Obomniru, G.E.; Oyadina, O.O.Background: Aloe vera gel (AVG) extract is commonly used as a home remedy for some diseases, including diabetes mellitus. Objectives: This study is to determine the effects of pure Aloe vera (Aloe babardensis) gel on some biochemical, haematological and histological parameters in diabetic rats. Methods: The rats were divided into six groups of six rats each. Diabetes mellitus was experimentally induced in rats by intraperitoneal administration of a single dose of alloxan monohydrate (170 mg/kg b.w.) following a 12 h fasting period. The Groups I and V received isotonic saline in a similar manner. The establishment of diabetes mellitus was confirmed by fasting blood glucose (FBG) level above 350mg/dl using a GlucoMetre (AccuChek Active). The AVG extract was administered for 14 days along with the metformin and alloxan drugs administered. Blood samples were taken from fasted rats. The effects of AVG on fasting blood, biochemical, haematological and oxidative stress parameters in the liver and histological examination of the liver, kidney and pancreas were evaluated. Results: There was a decrease in blood glucose level in the AVG treated group and an improvement in the lipid profile, as well as some haematological parameters. In addition, oral administration of Aloe barbadensis gel decreased the level of MDA with a corresponding increase in the activities of CAT, SOD, GST and GSH levels in the liver tissue of diabetic rats. Conclusion: These results suggest potent hypoglycaemic, hypolipidemic, antioxidant and therapeutic effects of Aloe Vera, beneficial as a herbal remedy for the treatment/management of diabetes mellitus.
